Recombinant production and characterization of Aspergillus niger prolyl endopeptidase enzyme for gluten-free food production

Gluten is a protein group found in wheat, barley, rye, and oats, known as cereals. When this vegetable introduced into the body, celiac disease can occurs. The use of bacterial fungal oligopeptidase to ensure cleavage gluten non-toxic fragments are considered promising alternative for disease. In study, Aspergillus niger Prolyl EndoPeptidase (AN-PEP) enzyme was cloned pET22b vector recombinantly produced BL21 (DE3) pLysE cells. PEP expressed inclusion body recovered by refolding. And N-terminal His-tagged recombinant purified nickel affinity chromatography. 280 mg AN-PEP from 1L culture at very high yield, 90% purity. As result; It has been determined that digest gluten. This study shows (rAN-PEP) great potential production processes gluten-free foods.

Production of the enzyme is decreased below pH 4.0 and is stimulated by the presence of substrate.
